General information

  • Activation of Wnt/β-catenin signaling occurs upon binding of Wnt to the 7-transmembrane domain–spanning frizzled receptor and low-density lipoprotein receptor–related protein 5 and 6 (LRP5/6) coreceptors. [1]
  • LRP5 was seen in Kupffer cells, particularly in those liver specimens with pathological evidence of hepatic congestion and in hepatic stellate cells. [2]
  • In the kidney, the convoluted tubules strongly expressed LRP5, whereas the glomeruli and collecting ducts showed no expression. [2]
  • Involved in the Wnt/beta catenin signaling pathway, probably by acting as a coreceptor together with Frizzled for Wnt. [3]
  • Mutations in the Wnt coreceptor lowdensity lipoprotein receptor–related protein 5 (LRP5) are causally linked to alterations in human bone mass. [1]
  • Wnt engagement of Lrp5 induces proliferation and extends the functional lifespan of osteoblasts. [4]
  • In addition to the beta-cells, there were isolated LRP5-expressing cells in the parenchyma of the pancreas which were identified as macrophages. [2]
  • LRP5/6 coreceptor activity is inhibited by members of the sclerostin (SOST gene product) and Dickkopf (Dkk) families, all of which bind LRP5/6. [1]

    Induced phenotypes

    • positive regulation of cell-matrix adhesion
      • Cultured rat hepatic stellate cells (HSCs) produce CTGF which is a cell adhesion factor for activated HSCs. LRP is a receptor for CTGF. [5]
      • LRP is utilized by HSCs for binding to CTGF and so this cell surface molecule may be involved in mediating CTGF activity or adhesive signaling during the activation process. [5]
