Details for receptor: MC1R

EndoNet ID: ENR00760

To link to the content of EndoNet use the EndoNet ID that is given on the detail pages in the format ENX0000, where X is a place holder for the type of the component (e. g. R for receptor or C for anatomical structure).
As URL for the linking append this ID to the detail page for this type of component.
For an hormone that would be:

http://endonet.bioinf.med.uni-goettingen.de/hormone/ENH00000

It is also possible to use the search of EndoNet to link to the right detail page. The URL should look like

http://endonet.bioinf.med.uni-goettingen.de/search/ENC00000
If the search pattern is unambigious the user is directed to the corresponding detail page.

General information

  • Regulates skin and hair pigmentation. [1]
  • MC1R is expressed by cutaneous melanocytes, where it has a key role in determining skin and hair pigmentation. [2]
  • Specific products were identified for MC1R in pituitary and testis. [3]
  • Membrane protein in melanocytes and corticoadrenal tissue. [1]
  • Coupled with G-protein that activates adenylyl cyclase. [1]
  • Receptor for MSH (alpha, beta and gamma) and ACTH. [1]

Binding hormones

  • melanotropin beta
    • The potency of MC1R-Ligands is alpha-MSH = ACTH > beta-MSH > gamma-MSH. [2]
  • melanotropin gamma
    • The potency of MC1R-Ligands is alpha-MSH = ACTH > beta-MSH > gamma-MSH. [2]
  • alpha-MSH
    • Activation of MC1R by alpha -MSH stimulates eumelanin synthesis in the melanocytes. [2]
    • MC1R is the "classical" melanocyte alpha -MSH receptor. [2]
    • The potency of MC1R-Ligands is alpha-MSH = ACTH > beta-MSH > gamma-MSH. [2]
  • ACTH
    • The potency of MC1R-Ligands is alpha-MSH = ACTH > beta-MSH > gamma-MSH. [2]

Anatomical structures with this receptor

  • melanocyte

    Influences

    • MSH

    Induced phenotypes

    • melanin synthesis
      • Alpha-MSH acts as an agonist by binding to melanocortin receptor 1 (MC1-R) and elevting intracellular cAMP to induce eumelanin synthesis in the follicular melanocytes. [4]
    • antiapoptotic effect
      • The antiapoptotic effect of alpha-MSH was linked to reduction in cyclopyrimidine dimers, the major UV-induced DNA photoproduct. The antiapoptotic effects of alpha-MSH and the reduction in UVB-induced cyclopyrimidine dimers were not observed in melanocytes expressing loss-of function mutation of MC-1R. [5]

    Induced phenotypes

    • suppression of interstitial fibrosis
      • Alpha-MSH suppressed interstitial fibrosis induced by TGF-beta1 and reduced the number of vimentin and alpha-smooth muscle actin positive cells. [6]
